Kisspeptin-10: what it is studied alongside
Kisspeptin-10 is a decapeptide fragment of the KISS1 gene product, studied as an agonist at the KISS1R receptor. This page summarises research on its co-administration with gonadotropin endpoints, its in vitro effects on
Kisspeptin-10 (also known as Kp-10, KiSS-10, or metastin 45–54) is a decapeptide corresponding to the C-terminal amidated fragment of the KISS1 gene product. It is classified as a neuropeptide agonist at the KISS1R receptor (formerly GPR54). The evidence block does not provide its sequence, molecular formula, or molecular weight; those identifiers are therefore not stated here. The work described below centres on the KISS1/KISS1R axis and its downstream effects on gonadotropin-releasing hormone (GnRH) and gonadotropin secretion.
Research on kisspeptin-10 spans reproductive neuroendocrinology, placental biology, metabolic regulation, and cardiac extracellular matrix remodelling. The sections that follow summarise findings from specific studies, with explicit notes where the evidence is animal-only or in vitro.
Research Studies
Kisspeptin-10 and gonadotropin secretion in healthy men
A randomised, single-blinded, placebo-controlled study evaluated acute and chronic subcutaneous kisspeptin-10 administration in healthy men. The protocol included an acute dose–response infusion, a 5-day continuous infusion, and daily intermittent administration (8 hours on, 16 hours off) for 12 days. The study reported that chronic subcutaneous kisspeptin-10 stimulated gonadotropin secretion for 12 days in this population [1].
Kisspeptin-10 and dynamic LH secretion during continuous infusion
A separate study examined the effects of continuous intravenous kisspeptin-10 infusion on luteinizing hormone (LH) secretion in healthy men. Three healthy adult men received a continuous infusion at 12.5 mcg/kg/h for 24 hours with frequent blood sampling. The objective was to characterise the effect on reproductive hormones, given that continuous kisspeptin administration had been shown to dampen kisspeptin-induced GnRH release in non-human primates but had not been replicated in the limited human studies reported to date [6].
Kisspeptin-10 and in vitro pharmacological comparison with analogs
An in vitro study compared the pharmacological profiles of hKp10, hKp54, and the synthetic agonist C6 in HEK293 cells. The kisspeptin analog C6 elicited greater tachyphylaxis and transcriptional activation than kisspeptin-10 and -54 [2]. This work was conducted in a cell line and does not establish in vivo potency or safety.
Kisspeptin-10 and glycosaminoglycan/decorin content in human cardiac fibroblasts
In human cardiac fibroblast cultures, kisspeptin-10 was studied for its influence on glycosaminoglycan and decorin content. The study assessed these effects with or without signalling inhibitors, measuring glycosaminoglycan levels by the Farndale method and decorin secretion by ELISA. The aim was to determine the influence of KiSS-10 on these extracellular matrix components and identify its mechanism [3]. This is an in vitro finding and does not demonstrate cardiac effects in whole organisms.
Kisspeptin-10 and neurokinin B/dynorphin A in ewe lambs
A study in ewe lambs examined changes in plasma concentrations of kisspeptin-10, neurokinin B (NKB), and dynorphin A (Dyn A) during puberty in relation to the first ovulation time. Twenty-four ewe lambs from single and twin pregnancies were divided into two groups. The aim was to determine the relationship between these neuropeptides during normal and delayed puberty [4]. This is an animal study; findings may not translate to humans.
Kisspeptin-10 and obesity-diabetes in high-fat fed female mice
In high-fat diet (HFD) fed female mice, twice-daily intraperitoneal administration of kisspeptin-10 (25 nmol/kg) for 21 days was examined for effects on glucose homeostasis, energy balance, circulating hormones, and the morphology–function of enteroendocrine and islet cells. Kisspeptin-10 decreased body weight, blood glucose, and energy intake to levels seen in normal diet (ND) mice. HFD increased circulating follicle-stimulating hormone (FSH) levels, which were further modulated by kisspeptin-10 [5]. This is an animal study and does not establish human metabolic outcomes.
Kisspeptin-10 and placental mTOR signalling in hypothyroid rats
In a rat model of maternal hypothyroidism induced by propylthiouracil, kisspeptin-10 treatment began on gestational day 8. The study evaluated maternal energy homeostasis and placental expression of mTOR and glucose metabolism mediators. The aim was to assess effects on placental mTOR signalling and glucose homeostasis disrupted by maternal hypothyroidism [8]. This is an animal study; placental findings in rats may not extrapolate to human pregnancy.
Kisspeptin-10 and the KISS1/KISS1R axis in human placentation
A review of the KISS1/KISS1R axis in human placentation discusses molecular mechanisms and implications for foetal growth restriction and pre-eclampsia. The review notes that kisspeptin and related biomarkers may improve early risk stratification and enable mechanistic classifications, as current diagnostic modalities predominantly reflect established disease rather than early molecular disturbances [7]. This is a review article and does not present new primary data.
Kisspeptin-10 and structural selectivity at KISS1R
A computational study reported structural insights into kisspeptin-10 selectivity toward KISS1R. The abstract notes that KP-10, the bioactive core fragment of kisspeptin neuropeptides, has emerged as a modulator of reward-related behaviours beyond its canonical role in reproductive neuroendocrinology, and that behavioural studies indicate KP-10 promotes impulsive and compulsive-like phenotypes in rodent models [9]. This is a reference-only item and is not the focus of this page.
Kisspeptin-10 and diagnostic ratios in precocious puberty
A study reported that the kisspeptin-10/basal LH ratio improves differentiation of central precocious puberty and premature thelarche. The background notes that distinguishing idiopathic central precocious puberty from premature thelarche remains a clinical challenge, often necessitating GnRH stimulation testing, and that kisspeptin-10, neurokinin B, and neuropeptide Y are key regulators of GnRH secretion that may serve as surrogate biomarkers [10]. This is a reference-only item.
Kisspeptin-10 and ovarian cancer cell lines
An in vitro and computational study explored the influence of exogenous kisspeptin-10 on the SP1-ZEB1-hTERT pathway in SKOV-3 ovarian cancer cell lines. The study aimed to investigate the effect on ovarian cancer progression through targeting this regulatory axis [11]. This is a reference-only item and does not establish clinical efficacy.
Kisspeptin-10 and ovarian follicle development in mice
A study in mice examined how KP-10 modulates hormone secretion, proliferation, and autophagy of ovarian follicles by regulating in vivo and in vitro levels of associated genes and proteins. Follicular granulosa cells were cultured in EMDM/F12 medium [12]. This is a reference-only item and an animal study.
