How to Reconstitute Kisspeptin-10
Kisspeptin-10 is a research peptide supplied as a lyophilised powder. This page covers its identifiers, the KISS1R/GnRH pathway, and the study models in which it has been examined.
Kisspeptin-10 (Kp-10, KP-10, KiSS-10) is the bioactive C-terminal decapeptide fragment of the kisspeptin neuropeptide family, derived from the KISS1 gene product. It is a research peptide supplied as a lyophilised powder for laboratory use. The evidence block for this page does not specify the amino acid sequence, molecular formula, or molecular weight of kisspeptin-10, so those values are not stated here. The work described below centres on the KISS1R receptor (also referred to as GPR54) and its downstream regulation of gonadotropin-releasing hormone (GnRH) release.
Reconstitution is the step of bringing a lyophilised peptide back into solution. The evidence block does not provide a reconstitution protocol, solvent recommendation, concentration, or storage condition for kisspeptin-10, so no such instructions are given on this page. What the evidence does provide is a body of study data on how kisspeptin-10 behaves in various models, which is summarised below.
Research Studies
Kisspeptin-10 and gonadotropin secretion in healthy men
A randomized, single-blinded, placebo-controlled study evaluated acute and chronic subcutaneous kisspeptin-10 administration in healthy men. The study characterised the acute dose-response relationship to subcutaneous kisspeptin-10 infusion, assessed continuous infusion over 5 days, and assessed daily intermittent administration (8 hours on, 16 hours off) for 12 days. Fifteen healthy men were recruited across three sub-studies, with 12 men serving as controls. Chronic subcutaneous kisspeptin-10 was reported to stimulate gonadotropin secretion for 12 days in this population [1].
Kisspeptin-10 and LH secretion during continuous infusion in healthy men
A separate study examined the effects of continuous kisspeptin administration on luteinizing hormone (LH) secretion in healthy men. Three healthy adult men received a continuous intravenous infusion of kisspeptin-10 at 12.5 mcg/kg/h for 24 hours and underwent frequent blood sampling. The authors noted that continuous kisspeptin administration had been shown to dampen kisspeptin-induced GnRH release in non-human primates, but that this had not been replicated in the limited human studies reported to date [6].
Kisspeptin-10 and the KISS1/KISS1R axis in human placentation
A review addressed the KISS1/KISS1R axis in human placentation, with implications for foetal growth restriction and pre-eclampsia. The review describes pre-eclampsia and foetal growth restriction as major pregnancy complications primarily driven by placental dysfunction, and discusses the identification of biomarkers directly associated with trophoblast signalling pathways [7].
Kisspeptin-10 and placental mTOR signalling in hypothyroid rats
An animal study evaluated the effects of maternal hypothyroidism, with and without kisspeptin-10 treatment, on maternal energy homeostasis and placental expression of mTOR and glucose metabolism mediators in rats. Maternal hypothyroidism was induced by propylthiouracil administration, and kisspeptin-10 treatment began on gestational day 8. The study was designed around the observation that maternal treatment with kisspeptin-10 had been shown to improve feto-placental development in hypothyroid rats [8].
Kisspeptin-10 and glycosaminoglycan and decorin content in human cardiac fibroblasts
An in vitro study determined the influence of kisspeptin-10 on glycosaminoglycan and decorin content in human cardiac fibroblast cultures and sought to identify its mechanism. Cardiac fibroblasts play a key role in extracellular matrix remodelling through synthesis of glycosaminoglycans and proteoglycans such as decorin. Glycosaminoglycan levels were determined by the Farndale method, and decorin secretion was measured, with and without signalling inhibitor [3].
Kisspeptin-10 and metabolic effects in high-fat fed female mice
An animal study examined the effects of twice-daily intraperitoneal administration of saline vehicle or kisspeptin-10 (25 nmol/kg) for 21 days on glucose homeostasis, energy balance, circulating hormones, and the morphology and function of enteroendocrine and islet cells in high-fat diet fed female mice, with normal diet mice as an additional control group. Kisspeptin-10 decreased body weight, blood glucose, and energy intake to normal diet levels. High-fat diet increased circulating follicle-stimulating hormone (FSH) levels, which were further altered by kisspeptin-10 [5].
Kisspeptin-10, neurokinin B, and dynorphin A in ewe lambs
An animal study determined changes in kisspeptin-10, neurokinin B (NKB), and dynorphin A (Dyn A) concentrations in blood plasma collected from ewe lambs during puberty in relation to first ovulation time, and examined the relationship between these neuropeptides. Twenty-four ewe lambs were divided into two groups: from single pregnancies and from twin pregnancies [4].
Kisspeptin-10 and the kisspeptin analog C6 in vitro
An in vitro study characterised and directly compared the pharmacological profiles of hKp10, hKp54, and the synthetic agonist C6 using HEK293 cells. The authors noted that direct comparative data on the effects of these molecules remain limited, both in vivo and in vitro, and that the kisspeptin analog C6 elicits greater tachyphylaxis and transcriptional activation than kisspeptin-10 and kisspeptin-54 [2].
Kisspeptin-10 in additional models
Additional findings from the evidence block are summarised below. These are reported as facts from the cited papers and are not the focus of any single section.
- A computational study reported structural insights into kisspeptin-10 selectivity toward KISS1R, describing kisspeptin-10 as the bioactive core fragment of kisspeptin neuropeptides and a modulator of reward-related behaviours beyond its canonical role in reproductive neuroendocrinology [9].
- A clinical study reported that the kisspeptin-10/basal LH ratio improves differentiation of central precocious puberty and premature thelarche, noting that distinguishing idiopathic central precocious puberty from premature thelarche remains a clinical challenge [10].
- An in vitro and computational study reported that exogenous kisspeptin-10 inhibits ovarian cancer progression through targeting the SP1-hTERT-ZEB1 regulatory axis in SKOV-3 ovarian cancer cell lines [11].
- An animal and in vitro study reported that kisspeptin-10 promotes hormone secretion, ovarian follicle development, and fecundity via the PI3K/AKT/ERK signal pathway in mice, with follicular granulosa cells cultured in EMDM/F12 medium [12].
