GHRP-6: What It Is Studied Alongside
GHRP-6 is a synthetic hexapeptide ghrelin-receptor agonist. This page maps the compounds, pathways and models it is studied alongside in the published literature.
Growth hormone-releasing peptide-6 (GHRP-6) is a synthetic hexapeptide growth hormone secretagogue and ghrelin receptor agonist. It is also written growth hormone releasing protein-6 and abbreviated GHRP-6; the evidence block for this page does not supply its sequence, molecular formula or molecular weight, so none are stated here. It is classed among the growth hormone secretagogues (GHS) alongside GHRP-2, hexarelin and ipamorelin, and among the peptides marketed as research compounds that modulate the GH-IGF-1 axis [5].
The work reviewed below centres on the ghrelin receptor (GHSR-1a), the receptor family through which GHRP-6 and ghrelin both act [3].
Research Studies
GHRP-6 and post-infarct ventricular remodelling in rats
In a permanent coronary ligation model, rats were assigned to sham-operated/normal, infarcted plus saline, and infarcted plus GHRP-6 groups, with treatment started after surgery and continued for 7 days. The authors had previously determined 0.4 mg/kg as the minimum effective dose for enhancing inotropy by echocardiographic parameters in healthy rats, and used a non-reperfusion myocardial infarct model with consequent left ventricle wall thinning and ballooning. GHRP-6 was reported to ameliorate post-infarct ventricular remodelling and systolic dysfunction in this model [1]. This is an animal study; no human cardiac endpoint is reported in the evidence.
GHRP-6 and endocrine and immune responses in gilthead seabream
GHRP-6 was included in aquafeed at 500 µg/kg of feed and assessed in Sparus aurata following Incomplete Freund's adjuvant (IFA) treatment. After 97 days, fish were injected intraperitoneally with 100 µL of saline solution or IFA per 100 g fish, and endocrine and immune responses were sampled. The study frames immunomodulatory feed additives as a strategy for fish health and performance in aquaculture [2]. This is a fish model; the findings are not transferable to mammals on the basis of this evidence alone.
GHRP-6 and locomotor activity during acute nicotine withdrawal in rats
Male and female Wistar rats received intraperitoneal nicotine at 2 mg/kg or saline twice daily for 7 days, and the study examined horizontal and vertical activity during acute withdrawal. Ghrelin and GHRP-6, which stimulate GH release through the same receptor, were tested for their impact on those withdrawal-associated changes in locomotor activity [3]. The work sits within a literature linking ghrelin and its receptor to reward sensation and addiction induced by natural and artificial drugs, including nicotine [3].
GHRP-6 and the ghrelin system in broiler chickens
Intracerebroventricular injection studies in broiler chickens examined liver-expressed antimicrobial peptide 2 (LEAP-2) and its interaction with cannabinoid and ghrelin systems on food intake. Across four experiments with four groups each, birds received saline, LEAP-2 at 0.75, 1.5 or 3 nmol, or the ghrelin receptor antagonist (D-Lys-3)-GHRP-6 at 0.5 nmol [6]. Here GHRP-6 appears as a pharmacological tool for probing ghrelin signalling rather than as the intervention of interest.
GHRP-6 and human oocyte maturation in vitro
An experimental study collected 240 human germinal vesicle (GV) oocytes and cultured them in varying concentrations of GHRP-6, described as a ghrelin hormone agonist, to determine the most effective concentration in in vitro maturation (IVM) culture medium. Maturation rates were assessed during culture [7]. This is an in vitro human cell study; it does not describe systemic administration or any in vivo endpoint.
GHRP-6 and the ghrelin-dopamine cascade in a rat model of depression
A novel butyrylcholinesterase inhibitor was evaluated in the Flinders Sensitive Line (FSL) rat model of major depressive disorder, with behavioural assessment by forced swim test, sucrose preference test and novel object recognition test. The paper situates its findings within a ghrelin-dopamine cascade [8]. GHRP-6 is not the administered agent in this study; the ghrelin system is the pathway context in which the results are interpreted.
GHRP-6 and acute lung injury progressing to fibrosis
Work in lipopolysaccharide (LPS) and zymosan (ZYM) induced lung injury biomodels, which recapitulate acute alveolar damage and progressive hypoxaemia, reports that GHRP-6 ameliorates acute lung injury and its subsequent evolvement to interstitial fibrosis [9]. This is a preclinical injury model.
GHRP-6 and acute kidney injury via metabolic regulation
Renal tubular epithelial cells are highly susceptible to injury during acute kidney injury and have notable regenerative effects on renal recovery. Because AKI-driven metabolic reprogramming of these cells helps determine whether kidneys recover functionally or develop fibrosis, GHRP-6 has been formulated in a hydrogel for acute kidney injury therapy via metabolic regulation [10]. This is preclinical formulation work.
GHRP-6 and nervous necrosis virus in fish
GHRP-6, described as a six-amino acid synthetic peptide acting as a ghrelin analog with growth-promoting and immunomodulatory activity in several fish species, has been evaluated against infection with nervous necrosis virus (NNV), an important pathogen in aquaculture [11]. This is a fish-pathogen model.

GHRP-6 and the wider GH-IGF-1 peptide landscape
A review of performance-enhancing peptides marketed as research compounds places GHRP-6 within the growth hormone secretagogue group, alongside GHRP-2, hexarelin and ipamorelin, and alongside GHRH analogues such as sermorelin, tesamorelin and CJC-1295 with and without DAC, the GH fragment AOD9604 (hGH 176-191), and IGF-1 analogues [5]. The same review notes that these agents are encountered in clinical practice and in online self-administration protocols [5].
GHRP-6 and ghrelin in reproductive and osteophytic contexts
Two further papers sit adjacent to GHRP-6 work without administering it. A case report describes a 62-year-old male ex-military recreational bodybuilder, a long-term growth hormone user, who presented with cervicalgia and left shoulder pain; imaging showed multilevel degenerative changes including a prominent anterior osteophyte at C3-C4 with esophageal compression, and he reported intermittent dysphagia with larger food boluses [4]. Separately, work on controlled ovarian hyperstimulation reports that COH increases circulating ghrelin and examines its role in COH-induced alterations [12]. Neither is a GHRP-6 study; both are cited here because they define the ghrelin-axis questions the peptide literature is read against.
