What is the half-life of semaglutide?
Semaglutide is dosed weekly in every registered trial in this pool. What that says about its half-life, and what half-life means for handling it in the laboratory.
Semaglutide is a glucagon-like peptide-1 receptor agonist, a synthetic analogue of the native incretin hormone modified to resist enzymatic breakdown and to bind albumin. Those modifications are the reason its half-life is long enough for the once-weekly dosing that every registered semaglutide trial in this pool uses [2][3][5].
Half-life is the time for the concentration of a compound to fall by half after it is administered. For a peptide it is set by three things: how fast enzymes cleave it, how fast the kidney clears it, and whether it is carried on a plasma protein. Native GLP-1 fails all three and lasts minutes; semaglutide is engineered to pass all three.
Research Studies
Semaglutide and weekly administration in registered trials
The trials registered on ClinicalTrials.gov in this pool — a placebo-controlled cardiometabolic study [2], a muscle-preservation study in older adults starting semaglutide [3], and a dose-effect study on hormone and craving outcomes [5] — all administer the compound once a week. A dosing interval that long is only possible for a molecule with a half-life measured in days.
Semaglutide and albumin binding
The fatty-acid side chain attached to the semaglutide sequence binds serum albumin, which shields the peptide from renal filtration and from enzymatic attack. This, rather than any change to receptor activity, is what extends the half-life from minutes to days.
Semaglutide and the steady state
A long half-life means the concentration climbs over several dosing intervals before it levels off. Trial protocols account for this with titration schedules; a laboratory account of exposure has to account for it too, because a single administration and a repeated one are not the same experiment.
Semaglutide and the half-life of the powder
Half-life in the body and shelf life in the vial are different quantities. The lyophilised material is stable as a dry cake; reconstituted solution is not, and should be aliquoted and kept cold. Research-grade semaglutide is supplied lyophilised with a per-batch analytical report.
Semaglutide and what this pool does not report
The registered trials in the evidence block record dosing schedules, not pharmacokinetic values. A specific half-life figure belongs to the pharmacokinetic literature and the product label, neither of which is in this pool, so none is quoted here.
Semaglutide and why native GLP-1 is short-lived
The native hormone is cleaved within minutes by dipeptidyl peptidase-4, which removes two residues from the N-terminus and inactivates it, and what escapes the enzyme is small enough to be filtered by the kidney. Semaglutide carries a substitution at the cleavage site that blocks the enzyme, a second substitution that supports the side-chain attachment, and the fatty-acid chain itself. Each modification addresses one of the three clearance routes; together they turn a minutes-long signal into a days-long one. This is also why the analogue and the native peptide, though they act at the same receptor, are not interchangeable in any experiment where duration matters.
Semaglutide and the incretin class
The same albumin-binding strategy underlies other weekly incretin agonists, and the registry lists dual-agonist studies alongside semaglutide ones [1]. The half-life engineering is a class property; the receptor profile is what differs.
