Common misconceptions about telomere length

Telomeres are repetitive DNA-protein structures at chromosome ends. This page examines what the research literature does and does not show about telomere length, with reference to Epitalon, collagen peptides, NMN, metfor

Telomeres are the repetitive DNA–protein structures that cap the ends of linear chromosomes; they are composed of tandem TTAGGG repeats bound by shelterin proteins. In the research literature, telomere length is treated as a biomarker of cellular aging, and the enzyme telomerase (encoded by TERT) is the primary mechanism by which telomeric repeats are extended. The work discussed here centres on telomerase activation and telomere maintenance pathways, including the alternative lengthening of telomeres (ALT) pathway.

Misconceptions about telomere length are common: that any intervention shown to lengthen telomeres in a cell line will do so in a whole organism, that telomere length is a direct measure of health, or that a single biomarker can stand in for the complex process of aging. The studies below examine specific compounds and mechanisms in defined models, and they should be read with those limits in mind.

Research Studies

Epitalon and telomerase upregulation or ALT activity in human cell lines

Epitalon, a naturally occurring tetrapeptide, has been studied for its effects on telomere length in human cell lines. The evidence indicates that Epitalon increases telomere length through telomerase upregulation or through ALT activity [4][5]. Quantitative data on the biomolecular pathway leading to telomere length increase, hTERT mRNA expression, telomerase enzyme activity, and ALT activation have been reported [4][5]. A correction to this work has also been published [3]. These findings are from human cell lines and do not establish effects in humans.

Collagen peptides and telomere length in adults aged 50–70

A clinical trial is investigating whether daily collagen peptide supplementation can stabilize or lengthen telomeres and improve related markers of cellular aging in adults aged 50–70 years with overweight and low-to-moderate physical activity [2]. The trial aims to determine whether six months of collagen peptides stabilizes or extends telomere length and increases telomerase activity compared with placebo, and whether any telomere-related changes are associated with lower inflammation, healthier body composition, and better functional health [2]. This is an ongoing study; no results are available.

Nicotinamide mononucleotide and immunosenescence in middle-aged and elderly individuals

A placebo-controlled clinical trial is investigating the effect of nicotinamide mononucleotide (NMN) supplementation on immunosenescence in middle-aged and elderly people with metabolic disorders [6]. The study aims to provide a new method to improve immunosenescence, and it examines effects on glucose and lipid metabolism [6]. Telomere length is not the primary endpoint of this trial; the focus is on immune and metabolic markers.

Metformin and molecular aging in prediabetes

Metformin has been shown to improve lifespan in animal models through an anti-ageing pathway driven by mTOR, and it has been shown to protect endothelial cells [7]. A clinical trial is examining metformin and molecular aging in individuals with prediabetes, a population with high rates of progression to type 2 diabetes [7]. The trial is ongoing; no results are available.

Transient reprogramming and endothelial cell aging

Non-genetic overexpression of the Yamanaka factors has been reported to induce partial functional rejuvenation and attenuation of senescence-associated features in endothelial cells [1]. The methods included quantification of gene expression and measurements of cellular functions in vitro, with further in vivo experiments performed in a hind-limb model [1]. This work is in endothelial cells and animal models; it does not address telomere length directly.

Telomere length and mitochondrial DNA copy number in porcine embryos

In parthenogenetically activated porcine embryos, mitochondrial DNA copy number and telomere length are considered secondary markers of embryonic quality [8]. The relationship between these markers has been investigated in this model [8]. This is an animal embryo study and does not inform human telomere biology directly.

Telomere length in patients with non-functional adrenal incidentalomas

Telomere length is affected by environmental factors such as oxidative stress and inflammation, in addition to genetic influences [9]. A study has examined telomere length in patients with non-functional adrenal incidentalomas, a condition linked to metabolic syndrome components and cellular aging [9]. This is an observational clinical study; it does not test an intervention.

Kisspeptin-54 and chondrocyte senescence in osteoarthritis

Kisspeptin-54, a neuropeptide, has been investigated for its role in chondrocyte senescence in osteoarthritis, with mechanisms involving SIRT3-mediated telomere protection and p53 acetylation inhibition [11]. This work is in chondrocytes and osteoarthritis models; it is not a human trial.

Breast DNA methylation-based measures of biological ageing and obesity-related risk

Epigenetic ageing measures have been linked to breast cancer risk, though prior studies have mainly focused on blood-based measures [10]. A study examined well-established epigenetic age estimates in normal breast tissue in relation to obesity-related breast cancer risk markers [10]. This is an observational study and does not address telomere length as an intervention target.
