Which peptides are studied in relation to skin elasticity?
A research-supply review of the peptides and peptide-containing formulations investigated in relation to skin elasticity, covering collagen peptides, acetyl hexapeptide-8, copper tripeptide, and multi-component nutraceut
Peptides studied in relation to skin elasticity fall into several distinct classes: signal and carrier peptides such as copper tripeptide, neurotransmitter-modulating peptides such as acetyl hexapeptide-8 (AH-8), hydrolysed and bioactive collagen peptides, and multi-component formulations in which peptides are combined with polyphenols, vitamins or other actives. The published and registered work spans in vitro assays, animal and ex vivo models, and human clinical trials, and the endpoints used to assess elasticity differ substantially between them.
Across this literature the mechanistic focus is the dermal extracellular matrix: elastin and collagen synthesis, elastase and matrix metalloproteinase activity, fibroblast function, and scaffold protein expression. Elastic fibre integrity is reported to decline with photoaging through increased elastase activity together with downregulation of elastin and scaffold-related gene expression, which is why multitarget approaches are studied rather than single-pathway interventions [2].
Research Studies
Copper tripeptide and multitarget elastic fibre restoration
An investigation of a multitarget approach to restoring the elastic fibre network examined modulation of elastin production, elastase activity and scaffold protein expression. The authors reported that copper tripeptide was among the agents studied in this context, with the rationale that preserving elastic fibre function requires addressing synthesis, enzymatic degradation and scaffold integrity simultaneously rather than elastin synthesis alone [2]. This work is mechanistic and does not by itself establish a clinical elasticity outcome.
Acetyl hexapeptide-8 and neuromuscular modulation
AH-8, often described as a botox-like peptide, is a biomimetic peptide used in topical cosmeceutical formulations intended for anti-aging, scar treatment and skin rejuvenation. A review of its skin permeability and efficacy describes its proposed mechanism as dynamic reduction of wrinkles through modulation of neuromuscular activity [6]. The review's focus is on structure, proposed mechanism and efficacy; it is a review rather than a primary elasticity trial.
Oral bioactive collagen peptides and skin properties in women with facial wrinkles
A randomised, double-blind, parallel-group trial enrolled 68 healthy women aged 35 to 65 years with noticeable facial wrinkles, allocated 1:1 to an active control or avvera. One participant in the active control group withdrew for personal reasons and was excluded from the final analysis, leaving 67 participants who completed the study. The trial evaluated whether daily oral intake of a collagen peptides supplement improved age-associated skin beauty parameters [7].
Collagen peptides and dermal fibroblast biology
A separate study evaluated a dietary supplement containing natural bioactive compounds, integrating in vitro and clinical approaches. Its stated rationale is that ageing involves progressive decline in dermal fibroblast function, with reduced proliferative capacity, decreased collagen synthesis and increased oxidative and glycative damage, leading to deterioration of skin elasticity and tissue homeostasis [5].
Ergothioneine and skin elasticity as a secondary endpoint
A randomised, double-blind, placebo-controlled trial is evaluating NVTIA Ergothioneine, a branded ergothioneine ingredient, in healthy adults aged 40 to 65 years. Participants receive the ingredient or matching placebo once daily for 12 weeks. The primary objective concerns biomarkers of oxidative stress and healthy aging; skin elasticity, skin hydration, inflammatory biomarkers and overall safety are secondary objectives [1]. Elasticity is therefore not the primary endpoint in this design.
Multi-component nutraceutical formulations and antioxidant status
A single-arm, open-label, prospective interventional study involving 55 participants evaluated NPQ Plus, a formulation containing polyphenols (Oligonol), nicotinamide mononucleotide (NMN; RevAge), pyrroloquinoline quinone (PQQ; PureQQ), coenzyme Q10 and a trace mineral complex (Ioniplex). Subjects consumed one sachet daily for 12 weeks, with assessments at baseline and at Weeks 4 and beyond, covering skin health, antioxidant status, fatigue and self-perceived well-being [4]. The design has no placebo comparator.
Hyaluronic acid injectable with L-carnitine and vitamin C
A post-market clinical investigation is evaluating Cellbooster Shape, a CE-marked hyaluronic acid-based superficial epidermis/dermis injection filler and skinbooster produced using CHAC Technology. It consists of non-crosslinked HA with L-carnitine and vitamin C and is designed to reduce local puffiness by improving microcirculation and hydration. Healthy subjects with signs of facial skin aging receive a 3-session treatment with follow-up over 70 days after the initial injection session, with several objective skin measurements recorded [3].
Oral capsule formulations and elasticity among exploratory outcomes
A 56-day, single-centre, open-label, single-arm exploratory study assessed within-participant changes after daily oral intake of NatureU Mind Care BeautyU Caps in adult women, one capsule daily for 56 days. The primary outcome was change from baseline in crow's-feet wrinkle count at Day 56 by PRIMOS CR imaging, and the secondary outcome was change in stratum corneum hydration by Corneometer CM 825. Skin elasticity was among the additional exploratory outcomes, alongside wrinkle topography, radiance, pigmentation, pore-related parameters, dermal thickness and density, and participant-reported outcomes [8].

Supporting in silico and in vitro work includes a knowledge-graph and molecular docking screen for cosmetic peptides with potential to enhance type III collagen, a collagen subtype described as playing a key role in maintaining skin elasticity and dermal resilience [10]. Related delivery research addresses the transdermal penetration barrier for hydrophilic low-molecular collagen peptides of approximately 5 kDa, using interface-engineered supramolecular collagen peptide nanocapsules for barrier repair and matrix remodelling [11]. An integrative in silico and in vitro evaluation of the pentapeptides Vialox and Leuphasyl examined antiaging potential in the context of extracellular matrix degradation, particularly of collagen and elastin [12]. A randomised controlled study of a comprehensive topical regimen incorporating niacinamide, Pro-Xylane and panthenol after PicoWay laser examined skin barrier repair and anti-aging effects [9].
