How Does Skin Elasticity Work?
Skin elasticity depends on the dermal elastic fiber network. This page outlines the mechanism and summarizes research on compounds studied for elastin synthesis, elastase suppression, and scaffold integrity.
How does skin elasticity work?
Skin elasticity is the mechanical property that allows skin to stretch and return to its original shape. It is primarily attributed to the elastic fiber network of the dermal extracellular matrix, which is composed of elastin and associated scaffold proteins. During skin aging, particularly photoaging, elastic fiber integrity is progressively compromised by increased elastase activity and the downregulation of elastin and scaffold-related gene expression [2].
The research summarized here centers on the elastin–elastase axis and the extracellular matrix (ECM) of the dermis, including collagen and elastic fiber components. The studies below are laboratory and clinical investigations of specific compounds; they are not intended as guidance for personal use.
Research Studies
Copper Tripeptide and Elastic Fiber Restoration in Dermal Models
A study investigating a multitarget approach to restoring the elastic fiber network examined the modulation of elastin production, elastase activity, and scaffold protein expression. The authors reported that Copper Tripeptide was among the agents evaluated for effects on elastin synthesis, elastase suppression, and scaffold reinforcement [2]. The work was conducted in dermal models and is not a human clinical trial.
Ergothioneine and Skin Elasticity in Healthy Adults
A randomized, double-blind, placebo-controlled clinical trial is evaluating NVTIA® Ergothioneine, a branded ergothioneine ingredient, in healthy adults aged 40 to 65 years. Participants receive either the active ingredient or a matching placebo once daily for 12 weeks. The primary objective is to assess biomarkers associated with oxidative stress and healthy aging; secondary objectives include assessment of skin elasticity, skin hydration, inflammatory biomarkers, and overall safety [1].
Cellbooster® Shape and Skin Aging Parameters in Healthy Subjects
A post-market clinical investigation is evaluating Cellbooster® Shape (CBS), a CE-marked hyaluronic acid-based superficial epidermis/dermis injection filler and skinbooster. CBS consists of non-crosslinked HA with L-carnitine and vitamin C and is designed for reducing local puffiness by improving microcirculation and hydration. The study enrolls healthy subjects with signs of skin aging in the face who receive a 3-session treatment and are followed for 70 days after the initial injection session. Objective measurements of skin parameters are included [3].
NPQ Plus and Skin Health in Healthy Adults
A single-arm, open-label, prospective interventional study evaluated NPQ Plus, a multi-component nutraceutical formulation containing polyphenols (Oligonol®), nicotinamide mononucleotide (NMN; RevAge®), pyrroloquinoline quinone (PQQ; PureQQ®), coenzyme Q10 (CoQ10), and a trace mineral complex (Ioniplex), in 55 healthy adults with skin concerns. Participants consumed one sachet daily for 12 weeks, with assessments at baseline and at Weeks 4, 8, and 12. Outcomes included skin health, antioxidant status, fatigue, and self-perceived well-being [4].
Dietary Supplement with Natural Bioactive Compounds and Collagen Production
A study integrating in vitro and clinical approaches evaluated a dietary supplement containing natural bioactive compounds on biological processes associated with skin ageing and extracellular matrix integrity. The rationale notes that ageing is characterized by a progressive decline in dermal fibroblast function, reduced proliferative capacity, decreased collagen synthesis, and increased accumulation of oxidative and glycative damage, leading to deterioration of skin elasticity and tissue homeostasis [5].
Acetyl Hexapeptide-8 and Skin Permeability in Cosmeceuticals
A review of acetyl hexapeptide-8 (AH-8), often referred to as a "botox-like" peptide, examined its structure, proposed mechanisms of action, skin permeability, and efficacy in topical formulations intended for anti-aging effects, scar treatment, and skin rejuvenation. AH-8 is described as a biomimetic peptide designed to mimic the function of naturally occurring peptides involved in skin homeostasis, repair, and regeneration, with potential to dynamically reduce wrinkles through modulation of neuromuscular activity [6].
Oral Bioactive Collagen Peptides and Skin Properties in Women
A randomized, double-blind, parallel-group clinical trial evaluated whether daily oral intake of a collagen peptides supplement improved age-associated skin beauty parameters in healthy women with visible facial wrinkles. A total of 68 healthy female volunteers between 35 and 65 years of age were initially enrolled and allocated 1:1 to receive either the active control (n = 34) or avvera™ (n = 34). One participant in the active control group withdrew, and 67 participants completed the study [7].
NatureU® Mind Care BeautyU Caps and Skin Aging Parameters
A 56-day, single-center, open-label, single-arm exploratory study evaluated within-participant changes in skin-aging parameters after daily oral intake of NatureU® Mind Care BeautyU Caps in adult women. Participants took one capsule daily for 56 days. The primary outcome was change from baseline in crow's-feet wrinkle count at Day 56 assessed by PRIMOS CR imaging; the secondary outcome was change from baseline in stratum corneum hydration at Day 56 assessed by Corneometer CM 825. Additional exploratory outcomes included other wrinkle topography parameters, skin elasticity, radiance, pigmentation, pore-related parameters, dermal thickness and density, and participant-reported skin assessments [8].
Topical Regimen with Niacinamide, Pro-Xylane, and Panthenol after Laser
A randomized controlled study evaluated a comprehensive topical regimen incorporating niacinamide, Pro-Xylane, and panthenol alongside other active agents after PicoWay laser. The study addressed whether the combination could deliver synergistic benefits for both skin barrier repair and anti-aging, noting that Pro-Xylane is a potent anti-aging ingredient but that limited knowledge exists regarding such post-procedure regimens [9].
Peptide Screening for Type III Collagen via Knowledge Graph and Molecular Docking
A study aimed to establish a data-driven approach to identify cosmetic peptides with potential to enhance type III collagen through multi-target mechanisms. Type III collagen plays a key role in maintaining skin elasticity and dermal resilience, and its decline is associated with visible signs of skin ageing [10].
Supramolecular Collagen Peptide Nanocapsules for Barrier Repair and Matrix Remodeling
Research on interface-engineered supramolecular collagen peptide nanocapsules addressed the transdermal delivery of hydrophilic low-molecular collagen peptides (MW ∼5 kDa), which remains a major dermatological barrier due to the strong barrier function of the stratum corneum. While recombinant collagen peptides hold therapeutic promise, their application is limited by poor penetration [11].
Vialox and Leuphasyl Pentapeptides in Antiaging Applications
An integrative in silico and in vitro study evaluated the antiaging potential of the cosmeceutical pentapeptides Vialox and Leuphasyl. The work notes that skin aging arises from intrinsic and extrinsic factors that drive extracellular matrix degradation, particularly of collagen and elastin, leading to wrinkles, pigmentation, and loss of skin elasticity [12].
