Which peptides are studied in relation to the senescence-associated secretory phenotype?
A review of research on peptides and related compounds studied in the context of the senescence-associated secretory phenotype (SASP), including SS-31, piperlongumine, and senolytic approaches.
The senescence-associated secretory phenotype (SASP) is a hallmark of senescent cells, characterized by the secretion of inflammatory cytokines, chemokines, growth factors, and matrix-remodeling enzymes that alter the tissue microenvironment [2]. In research settings, several peptides and small molecules have been investigated for their ability to modulate SASP or eliminate senescent cells. This page summarizes studies on peptides such as SS-31 (elamipretide) and other agents like piperlongumine, as well as senolytic strategies, with a focus on the mechanisms and models used.
The work centres on pathways including NF-κB, mTOR, JAK/STAT, and cGAS-STING, which are key signaling cascades implicated in SASP regulation [4].
Research Studies
SS-31 and radiation-induced cardiomyocyte senescence
SS-31 (elamipretide) is a mitochondrial-targeted peptide that was evaluated for its effects on radiation-induced senescence in cardiomyoblast H9C2 cells and human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). Exposure to γ-radiation at doses of 2, 5, and 10 Gy inhibited H9C2 cell proliferation in a dose-dependent manner and induced senescence-associated beta-galactosidase (SA-β-gal) staining. Treatment with SS-31 attenuated these senescence markers in vitro [6]. This study was conducted in cell models, not in animals or humans.
Piperlongumine and senescent meniscus cells and chondrocytes in osteoarthritis
Piperlongumine (PL), a small molecule, was investigated for its senolytic effects in the context of knee osteoarthritis. In a rat model of OA, senescence and SASP marker expression were assessed in meniscus and cartilage. Rat meniscus cells and chondrocytes were rendered senescent with H2O2 and treated with candidate senolytics, including PL. Transcriptomic analysis explored the mechanism, and PL was found to selectively eliminate senescent cells [7]. This research was conducted in vitro and in animal models.
Senolytic-regenerative therapy and healthspan in murine models
A combination approach using SenoVax™, a dendritic cell-based senolytic immunotherapy, combined with syngeneic pluripotent stem cell-derived mesenchymal stem cells (MSCs), was tested in murine models of hepatic injury and accelerated aging. The treatment aimed to reduce senescent cell burden and counteract the anti-regenerative effects of SASP factors. The study reported synergistic effects on healthspan and lifespan in mice [8]. This evidence is animal-only.
Senomorphic agents and SASP modulation
Senomorphic agents are compounds that modulate SASP without eliminating senescent cells. A review classified these agents into three categories and described their multi-target mechanisms, including inhibition of NF-κB, mTOR, JAK/STAT, and cGAS-STING pathways [4]. This work is a review of preclinical and clinical evidence, not a primary study.
CCT/TRiC chaperonin and therapy-induced senescence in glioblastoma
In glioblastoma multiforme (GBM), standard therapies can induce therapy-induced senescence (TIS) marked by SASP. The eukaryotic chaperonin CCT/TRiC, a proteostasis network component, has been studied in this context. GBM is chaperone-dependent, and CCT/TRiC may influence TIS and SASP [3]. This research is based on cell and molecular studies, not clinical trials.
Circulating biomarkers of senescence and therapeutic interventions
A review consolidated human-focused evidence on circulating biomarkers of senescence, including SASP factors, and discussed emerging drug and lifestyle-based senotherapeutic interventions [5]. This work is a review of human observational and interventional studies.
Bone aging and multiscale degeneration
Bone aging is characterized by declines in bone mass, microstructure, and biomechanical properties, and involves molecular, cellular, tissue, and system levels. SASP is implicated in this process [1]. This review synthesizes evidence from various models, including human and animal studies.
Radiation-induced atrial fibrillation and senolytic therapy
Radiation-induced atrial fibrillation is driven by cellular senescence via the GATA4-NF-κB pathway and was rescued by D/Q senolytic therapy in preclinical models [9]. This finding is from animal and in vitro studies.
Intracranial aneurysms and vascular aging
Intracranial aneurysms are associated with cellular senescence, inflammaging, and wall degeneration, suggesting a role for SASP in premature vascular aging [10]. This evidence is from human and animal studies.
Chronic intracellular infection and senescence
Mycobacterium abscessus reprograms alveolar macrophages towards a senescent state, highlighting a role for senescence in chronic infection [11]. This research is based on in vitro and animal models.
Ovarian aging and systemic health
Ovarian aging may contribute to systemic aging via SASP-related inflammaging and fibrosis [12]. This review discusses mechanisms and emerging interventions, drawing from human and animal studies.
