Oxytocin acetate: what it is studied alongside
Oxytocin acetate is the acetate salt of the nonapeptide oxytocin, studied in laboratory and clinical research alongside estrogen-progestin provocation, prostaglandin F2α, scopolamine, and oxytocin/vasopressin receptor an
Oxytocin acetate is the acetate salt form of oxytocin, a nine-amino-acid cyclic nonapeptide of the posterior pituitary hormone class. Synonyms encountered in the literature include oxytocin diacetate and oxytocin triacetate, reflecting variable acetate stoichiometry in the salt form. The parent peptide has the sequence Cys-Tyr-Ile-Gln-Asn-Cys-Pro-Leu-Gly-NH2 with a disulfide bridge between the two cysteine residues, a molecular formula of C43H66N12O12S2, and a molecular weight of approximately 1007.19 g/mol; the acetate salt adds mass according to the number of acetate counterions present. Identifiers commonly attached to the parent compound include CAS 50-56-6 and PubChem CID 439302. Oxytocin acetate is supplied as a research reagent for laboratory use.
The work around this compound centres on the oxytocin receptor, a class A G protein-coupled receptor that also binds vasopressin and is the target through which oxytocin and its synthetic analogues exert their measured effects.
Research Studies
Oxytocin acetate and uterine involution in postpartum dairy cows
A comparative study in cattle examined three substances with ecbolic activity — oxytocin, prostaglandin F2α, and scopolamine — for their effect on uterine involution and the resumption of ovarian activity after calving. Eighty bovine animals were randomly assigned to four groups of twenty. One group received saline within 24 hours of calving, one received 150 µg/head of d-cloprostenol, one received 50 IU/head of oxytocin acetate, and one received 40 mg of scopolamine butylbromide. Blood sampling was used to evaluate hydroxyproline and related parameters [3]. This is an animal study; findings in dairy cows do not translate directly to other species or to human physiology.
| Group | Treatment within 24 h of calving | n |
|---|---|---|
| C | 5 mL/head saline solution | 20 |
| PG | 150 µg/head d-cloprostenol | 20 |
| OX | 50 IU/head oxytocin acetate | 20 |
| S | 40 mg scopolamine butylbromide | 20 |
Oxytocin and estrogen-progestin provocation in adults with pituitary disease
An open-label pilot study is characterising the oxytocin response to a single oral dose of estrogen-progestin in adults with arginine-vasopressin deficiency compared with healthy controls. The protocol also examines associations between oxytocin levels and measures of psychopathology — anxiety and depression — and quality of life across groups. The stated hypotheses are that salivary and blood oxytocin responses to estrogen-progestin will be lower in arginine-vasopressin deficiency than in healthy controls, and that lower salivary and blood oxytocin levels will be associated with more severe anxiety, depression, and social-emotional difficulties as well as lower quality of life [2]. This is a registered protocol; results are not reported in the evidence available here.
Oxytocin and estrogen-progestin provocation in pediatric pituitary disease
A separate open-label interventional pilot trial is recruiting ten youth with arginine-vasopressin deficiency and ten with pituitary disease, matched for age, sex, and BMI, from pediatric endocrinology and neuroendocrinology clinics and from the community. The study tests the hypothesis that oral estrogen-progestin will stimulate endogenous oxytocin release in control subjects. Eligible participants receive two tablets in a single administration containing a total of 1 mg norethindrone acetate and 70 µg ethinyl estradiol. Blood and saliva are sampled at baseline and approximately 24 hours after study drug administration, alongside neuropsychological assessment of anxiety, mood, and emotion regulation [1]. As with the adult protocol, this is a registered study design rather than a reported outcome.
Oxytocin receptor antagonism and preterm labour: atosiban and retosiban
A randomized, double-blind, double-dummy study compared retosiban with atosiban in women in spontaneous preterm labour. The primary objective was to demonstrate superiority of retosiban in prolonging pregnancy. The design rests on the hypothesis that prolonging time to delivery in the absence of harm may benefit the newborn, particularly in women presenting at early gestational ages. Atosiban is described in the protocol as a mixed oxytocin-vasopressin antagonist indicated for short-term use to delay imminent preterm birth in women between 24+0/7 and 33+6/7 weeks' gestation [7]. This places oxytocin receptor pharmacology alongside a direct antagonist comparator in an obstetric setting.
Oxytocin and cardiac output during cesarean delivery under spinal anesthesia
A study protocol describes measurement of cardiac output in healthy parturients undergoing cesarean delivery under spinal anesthesia [4]. The evidence available here does not specify the oxytocin exposure or comparator used in that protocol, so no claim about an oxytocin effect on cardiac output can be drawn from it.
Oxytocin-adjacent obstetric research: fluid assessment in preeclampsia
A separate protocol addresses ultrasonography for fluid assessment in parturients with preeclampsia undergoing cesarean section. Preeclampsia is described as a multifocal syndrome reported in 2–8% of pregnancies, diagnosed in the second half of pregnancy by two separate measurements of systolic blood pressure ≥140 mmHg and/or diastolic blood pressure ≥90 mmHg in the same arm, together with proteinuria >300 mg in a 24-hour urine collection. The risk of serious complications such as pulmonary edema, cerebrovascular accidents, coagulopathy, and hemorrhage is reported as 10- to 30-fold higher among parturients with severe preeclampsia, defined by severe hypertension (systolic ≥160 mmHg and/or diastolic ≥110 mmHg on more than one occasion) or other listed features [5]. This protocol does not itself test oxytocin; it is included here because it defines the obstetric population in which oxytocin-related haemodynamic and fluid questions are frequently posed.
Oxytocin-adjacent obstetric research: colloid and crystalloid comparison
A further protocol compares a combination of colloids and crystalloids with crystalloids alone in women with preeclampsia undergoing elective cesarean delivery under spinal anesthesia [6]. As with the preceding entry, the evidence available does not report an oxytocin arm, and no oxytocin-specific conclusion should be drawn from it.
Oxytocin receptor agonism combined with rTMS in a postpartum depression model
Work in mice has examined a combination of repetitive transcranial magnetic stimulation and an oxytocin agonist in relation to depression-like behaviour after postpartum depression. The paper situates postpartum depression within DSM-5 as a subtype of major depressive disorder with peripartum onset, generally arising within the initial trimester following delivery, and characterised by acute psychiatric features [8]. This is an animal model; the findings are not evidence of an effect in humans.
