What Is Intestinal Permeability?
Intestinal permeability describes the regulated passage of solutes across the gut epithelium. This page covers the assays, models and research compounds used to study it, and states plainly where the evidence is animal-o
Intestinal permeability is the property of the intestinal epithelium that governs the rate and selectivity with which solutes cross from the lumen into the subepithelial space. It is not a single molecule or receptor but a functional characteristic of a tissue, and it is measured rather than defined by any one identifier: no sequence, formula or molecular weight attaches to it. In the research literature the term is used interchangeably with gut barrier function, and the two are frequently paired with the phrase "leaky gut" in non-specialist writing. The measurement that recurs across the studies cited here is the urinary lactulose : mannitol ratio, collected over a 5-hour period, which reports permeability as a ratio rather than an absolute flux [6].
The work centres on the paracellular route through the tight-junction protein family that seals adjacent enterocytes, with transepithelial transport across the monolayer as the complementary pathway; the two together determine what reaches the lamina propria and, subsequently, the circulation.
Research Studies
Marine collagen peptides and barrier integrity in Caco-2 and murine colitis
A low-molecular-weight collagen peptide fraction designated LSCP-I, derived from the scales of Lutjanus erythropterus and recovered as a <3 kDa fraction from food-processing byproducts, was evaluated in cellular and murine models of colitis. In Caco-2 cells subjected to macrophage-mediated inflammatory injury, LSCP-I at 50 µg/mL increased proliferation by 35% and migration by 40%, preserved barrier integrity, reduced reactive oxygen species by 45%, decreased lipid peroxidation by 30%, and restored glutathione and superoxide dismutase [1]. These are in vitro observations in a human colorectal adenocarcinoma cell line, and the accompanying colitis work was performed in mice; neither establishes an effect in humans.
Oral glutamine and sex-dependent gut responses in diet-induced obesity
Male and female C57BL/6 mice received a standard diet or a high-fat diet providing 60% of kcal from fat for 14 weeks. From week 12 onward, animals received glutamine in drinking water at 2 g/kg/day or no supplementation, and body composition, glucose tolerance, insulin sensitivity, intestinal permeability and colon outcomes were characterised [2]. The study's framing is that obesity is associated with sex-dependent metabolic complications to which altered intestinal barrier function and gut microbiota contribute, and it set out to compare male and female responses to the same oral glutamine exposure. This is a mouse study; the sex-divergent findings reported in it are findings in mice.
Zein-based nanoparticles and oral peptide permeability in Danio rerio
Hybrid zein-Eudragit RS100-chitosan nanoparticles were developed for the co-delivery of insulin and liraglutide, with the stated aim of protecting both peptides and supporting oral administration against enzymatic degradation and poor intestinal permeability. The particles were prepared by nanoprecipitation and exhibited nanoscale size of approximately 230 nm, polydispersity below 0.1, and a positive zeta potential of approximately +46 mV, with FTIR, XRD, TGA and DSC indicating a predominantly amorphous and molecularly dispersed structure [3]. Preliminary in vivo evaluation was conducted in Danio rerio; the formulation and transport findings are not human data.
Collagen peptide dose-ranging against aspirin challenge in healthy adults
A registered trial is evaluating 45 days of daily collagen peptide administration at three doses — 2.5 g, 5 g and 10 g — against placebo in healthy adults, with intestinal permeability following an acute aspirin challenge as the outcome. The primary comparison is change from Day 42 to Day 45 after the NSAID challenge, measured by the urinary lactulose : mannitol ratio over a 5-hour collection [6]. This is a registered protocol; the design is reported here, not results.
Inulin and pectin fibre in alcohol use disorder
A registered trial in patients with alcohol use disorder aged 19 to 65 is testing whether a fibre-enriched diet with a high proportion of inulin and pectin, combined with standard care, reduces intestinal permeability. The stated hypothesis is that a diet rich in these fibres modifies the gut microbiota and its metabolites, inducing a decrease in intestinal permeability, restoring microbiota composition and metabolites, and further improving abstinence, craving and anxiety, inflammation, steatosis and hepatic fibrosis [7]. The trial is registered; no outcome data are reported in the record cited.
Sugar-based synbiotic, inulin and Bacillus coagulans BC30 in overweight adults
A randomised, double-blind, controlled crossover study is investigating a sugar-based synbiotic containing inulin and Bacillus coagulans BC30 spores in healthy overweight individuals. Twenty participants complete a 2-week run-in and two 4-week intervention periods separated by a 4-week washout, consuming two sachets daily of either the synbiotic, providing 1 × 10⁹ CFU/day of BC30, or a matched control sugar product containing inulin without probiotic spores. The primary outcome is change in fasting markers of subclinical inflammation, including high-sensitivity C-reactive protein [5]. Intestinal permeability is not the primary endpoint in this record.
Akkermansia muciniphila and berberine in night-shift workers
A registered study addresses night shift work, which is associated with increased risk of obesity, insulin resistance and cardiometabolic disorders, largely through circadian misalignment, disrupted sleep and altered eating patterns. Akkermansia muciniphila has been linked to improved metabolic health including enhanced insulin sensitivity, lipid regulation and maintenance of intestinal barrier integrity, and berberine, a plant-derived compound, has shown metabolic effects including upregulation of A. muciniphila and improvement of insulin sensitivity [8]. The record describes the rationale and design; it does not report permeability outcomes.
Rowing exercise and multilevel intestinal response
An observational study of thirty members of the Polish Youth National Rowing Team, aged 19 to 24, is designed to evaluate multilevel physiological, molecular, metabolic, intestinal, immunological and psychophysiological responses to rowing-specific exercise across two training periods. It aims to investigate how maximal and prolonged rowing ergometer exercise, including a 2000-m maximal effort during the competitive phase, influences integrated adaptive mechanisms related to mitochondrial function, metabolic regulation, intestinal permeability, immune activation, DNA damage response and psychological status [4]. This is a registered observational protocol in athletes.
Permeation enhancers and PROTAC transport in rats
Work on the proteolysis targeting chimera ARV-471 examined the effects of intestinal permeation enhancers on solubilisation and epithelial transport in rats. The reported context is that PROTAC molecular properties often fall outside the rule-of-five drug-like chemical space, which is associated with poor intestinal solubility, permeability and absorption, and that formulation strategies have focused primarily on solubility enhancement while intrinsic permeability remains a constraint [9].
Natural products and NSAID-induced small intestinal injury
A systematic review evaluated natural products, functional foods and probiotics for preventing or treating NSAID-induced small intestinal injury, a complication among chronic NSAID and aspirin users for which effective treatment options remain limited, with mechanistic consideration given to oxidative stress and microbiome modulation [10].
Canagliflozin and gut integrity in indomethacin-induced ulcer
Preclinical work assessed the gastroprotective potential of canagliflozin in indomethacin-induced peptic ulcer, with reported impact on inflammation, pyroptosis, autophagy and gut integrity. The framing is that indomethacin is a common analgesic and anti-inflammatory agent whose prolonged use is linked to gastrointestinal bleeding, ulcers and perforations [12].
Permeability-adjacent biomarkers in PCOS
A review of diagnostic and prognostic biomarkers in polycystic ovary syndrome examines insulin resistance, systemic inflammation and endometrial receptivity, describing insulin resistance as a central molecular driver of metabolic and reproductive complications including infertility, obesity and increased long-term cardiovascular risk [11].
