Which peptides are studied in relation to the extracellular matrix?

A research-supply overview of peptides and ECM-associated proteins examined in extracellular matrix studies, spanning periostin, fibronectin-binding peptides, BDNF, MAFF and related targets in fibrosis, bone, muscle and

Peptides studied in relation to the extracellular matrix (ECM) are a heterogeneous set: some are ECM structural or matricellular proteins themselves, some are short synthetic binding peptides used as probes of matrix mechanics, and some are signalling proteins whose expression is measured alongside matrix remodelling. The evidence summarised here covers periostin (POSTN), a multifunctional ECM protein; the fibronectin-binding peptide FnBPA5; brain-derived neurotrophic factor (BDNF); the transcription factor MAFF; and matrix-associated targets discussed in bone-aging, cardiac-fibrosis and cosmetic-science reviews. Where the evidence gives identifiers, they are reported as stated; where it does not, none are supplied.

The work centres on integrin-linked and growth-factor-linked signalling to matrix-producing cells — integrin–JNK cascades, TGFβ-driven fibronectin assembly, and BET-protein-dependent fibrotic activation — together with matrix metalloproteinases and CD44 as targets in skin and connective-tissue research [2][3][7][12].

Research Studies

Periostin (POSTN) and cardiac fibrosis in arrhythmogenic cardiomyopathy models

POSTN is described as a multifunctional extracellular matrix protein in cardiovascular disease. In arrhythmogenic cardiomyopathy (ACM), POSTN was significantly elevated in the myocardium of ACM mice and of patients. POSTN deficiency alleviated heart dysfunction, reduced cardiac fibrosis and improved survival in ACM mice. The protein was mainly derived from cardiac myofibroblasts and promoted cardiomyocyte necroptosis in vivo and in vitro. Mechanistically, POSTN facilitated necroptosis through integrin–JNK–RIP3–MLKL signalling and induced CCL3 expression via an integrin–JNK–ETS2 pathway; CCL3 in turn activated cardiac myofibroblasts, forming a feedforward loop [2].

FnBPA5 and fibronectin tension in idiopathic pulmonary fibrosis models

FnBPA5 is a tension-sensitive fibronectin-binding peptide whose multivalent binding to fibronectin is impaired by fibre strain. An in vitro fibrosis assay using patient-derived fibroblasts and FnBPA5 was established to test the relationship between ECM fibre tension and fibrosis progression in idiopathic pulmonary fibrosis (IPF); the system was validated against known fibrosis biomarkers at protein and mRNA levels [8].

Apabetalone and fibrotic activation of cardiac fibroblasts

Apabetalone is a clinical-stage BET inhibitor. It was tested for anti-fibrotic activity in cell models recapitulating cardiac fibrosis, including human immortalised cardiac fibroblasts, cryopreserved cardiac fibroblasts, and ex vivo cultured fibroblasts from patients undergoing bypass grafting. The study addressed ECM accumulation as a driver of myocardial dysfunction and heart failure, noting that no approved anti-fibrotic therapeutics target it [3].

BDNF and intervertebral disc degeneration

BDNF is a neurotrophin family protein involved in neuronal survival, growth and synaptic plasticity. BDNF expression has been confirmed within intervertebral disc tissue, with degenerated discs showing increased expression compared with normal discs, and expression reported to increase further with degeneration severity. Disc degeneration is closely associated with intradiscal nerve ingrowth. A registered study examines correlation between serum BDNF levels, Pfirrmann grading and IL-1β [5].

MAFF and contraction-induced transcription in skeletal muscle

MAFF is a small MAF transcription factor consistently induced by acute exercise in both human and mouse skeletal muscle, with induction restricted to muscles recruited by the exercise modality. In C2C12 cells, Maff was induced by in vitro electrical pulse stimulation. CRISPR/Cas9-mediated Maff knockout C2C12 cells showed reduced myogenic fusion with accumulation of unfused nuclei, and RNA sequencing revealed broad reprogramming in Maff-deficient myotubes [4].

Fibronectin matrix assembly in astrocytes versus fibroblasts

Fibrillar fibronectin matrix is the predominant ECM in most tissues, but in neural tissues it is limited to sites of tissue damage and disease. Primary cortical astrocytes were shown to produce fibronectin and assemble it into fibrils through a process that differs from fibroblast behaviour under TGFβ [12].

Matrix targets in bone aging research

Bone aging is characterised not only by bone mass loss but as a multi-level degenerative phenomenon spanning molecular, cellular, tissue and system levels, with declines in bone mass, microstructure and biomechanical properties [1].

Matrix metalloproteinases and CD44 in cosmetic science

A review of molecular targets in cosmetic research highlights matrix metalloproteinases, the CD44 receptor, telomerase, histone deacetylases (particularly sirtuins), antioxidant enzymes, tyrosinase, MITF and melanocortin receptors as regulators of DNA repair, gene expression and ECM-related processes in skin aging, pigmentation, inflammation and tissue regeneration [7].

ECM protein interactions at implant interfaces

Graphene oxide–hydroxyapatite nanocomposite coatings were examined for ECM protein interactions relevant to osseointegration in dental implants. Molecular docking, pharmacophore modelling and molecular dynamics simulations explored interactions of GO, HA and GO-HA composites; GO has strong protein-binding ability attributed to surface area and oxygen functional groups but dose-dependent cytotoxicity, while HA is biocompatible with weaker protein–protein interactions at the molecular scale [6].
