What does the research say about elastin?

Elastin is the extracellular matrix protein that gives vertebrate connective tissue its reversible extensibility. This page summarises published research on elastin, solubilised elastin peptides, and elastin-derived biom

Elastin is the extracellular matrix (ECM) protein responsible for the reversible extensibility of vertebrate connective tissue, and it is the principal structural component of elastic fibres alongside fibrillin-rich microfibrils. In the research literature it appears under the names elastin, tropoelastin (the soluble monomeric precursor), and, for the degradation products, elastin-derived peptides (EDPs) or elastin peptides (EPs). The evidence block supplied for this page does not give a sequence, formula or molecular weight for elastin itself; the only molecular-weight information present concerns solubilised elastin preparations, where the stepwise oxalic-acid-hydrolysed material (OxA-st-ELN) was reported to have the largest fragment molecular weights among five preparations compared [5].

The work summarised here centres on the elastin receptor complex and on elastase-mediated turnover of the elastic fibre: studies describe elastin peptides interacting with elastin receptor complexes, and describe elastic fibre integrity as a balance between elastin and scaffold gene expression on one side and elastase activity on the other [2][6].

Research Studies

Elastin peptides and glucose metabolism in stroke-prone spontaneously hypertensive rats

Bonito bulbus arteriosus-derived elastin peptides were administered orally to male stroke-prone spontaneously hypertensive rats (SHRSP) with glucose intolerance, either as a single 1000 mg/kg dose or as a 4-week regimen at 600 mg/kg/day. The study measured glucose tolerance, plasma GLP-1 and insulin levels, and renal vascular integrity, and reported improved glucose metabolism and protection of the renal vasculature alongside modest dipeptidyl peptidase 4 inhibition [1]. This is an animal model; the findings are not human data.

Elastin upregulation, elastase suppression and scaffold reinforcement in dermal elastic fibres

A multitarget approach to restoring the dermal elastic fibre network was investigated by modulating elastin production, elastase activity and scaffold protein expression. The authors frame photoaging as a process in which elastic fibre integrity is progressively compromised by increased elastase activity and downregulation of elastin and scaffold-related gene expression, and argue that preserving elastic fibre function requires addressing synthesis, enzymatic degradation and scaffold integrity together [2].

Elastin-derived peptide hydrogels and tetrapeptide-21 delivery

First- and second-generation elastin-derived peptides were designed, synthesised and evaluated as hydrogel-forming carriers for the collagen-stimulating tetrapeptide-21 (GEKG), on the basis of self-assembly, crosslinking ability and sustained bioactive peptide release. Transmission electron microscopy showed uniform nanofibres with a hollow structure, and encapsulation and permeation properties were assessed [3]. This is a delivery-system study rather than a study of elastin as a therapeutic agent.

Solubilised elastin preparations and cellular response in fibroblasts and macrophages

Five solubilised elastins were prepared by chemical, enzymatic or combined hydrolysis: stepwise and continuously oxalic-acid-hydrolysed (OxA-st-ELN, OxA-ELN), potassium hydroxide-hydrolysed (KOH-ELN), enzymatically hydrolysed (Enz-ELN), and combined (Combi-ELN). OxA-st-ELN had the largest molecular-weight fragments, and cellular responses were compared on fibroblasts and macrophages [5]. The paper notes that chemically solubilised elastins have shown advantageous effects in skin regeneration in humans, and that solubilised elastin peptides retain elastin bioactivity and are considered more suitable for wound care than the insoluble form [5].

Elastin peptides and elastic fibre injury in pulmonary emphysema models

Degradation of elastic fibres is a characteristic of pulmonary emphysema and releases proinflammatory elastin peptides. In a lipopolysaccharide-induced model of acute lung injury, intratracheal instillation of elastin peptides significantly enhanced breakdown of these fibres, a result consistent with a mechanism in which an expanding pool of elastin peptides generates further elastic fibre injury through interaction with elastin receptor complexes [6]. This is an animal model and a hypothesis-driven review of laboratory findings.

Elastin and collagen alterations in ligamentum flavum hypertrophy

An ongoing study is analysing proteomic and histological changes in ligamentum flavum samples from patients with lumbar spinal stenosis undergoing surgery, correlating molecular signatures with imaging and surgical findings. The background states that fibrotic processes involving elastin and collagen alterations contribute to ligamentum flavum thickening and spinal instability, and that the molecular mechanisms remain unclear [4]. This is a registered study record; results are not reported in the evidence block.

Elastin production as an outcome measure in facial scar studies

A registered trial of botulinum toxin A and mesotherapy for facial scarring describes mesotherapy as a non-invasive transdermal injection that can aid the skin to increase collagen and elastin production, and notes that botulinum toxin A is known to prevent fibroblast proliferation and to induce temporary muscle paralysis [7]. Elastin production is referenced as a rationale rather than reported as a measured endpoint in the evidence block.

Elastin in gingival recession grafting comparisons

A registered trial compares a volume-stable collagen matrix of porcine origin with subepithelial connective tissue graft combined with a coronally advanced flap in RT2 gingival recession. The record notes that significant resorption of connective tissue graft material has been reported when the graft is exposed, which can reduce the possibility of complete root coverage [8]. Elastin is not the intervention in this study.

Elastin-containing and elastin-like materials in tissue engineering and matrix biology

Several reference items describe elastin-adjacent materials rather than elastin itself. A retraction notice concerns a gelatin-elastin-hyaluronic acid hydrogel scaffold with pectin microparticles for bone tissue engineering [9]. A chitosan hydrogel loaded with mesenchymal stem cell-derived exosomes accelerated wound closure and enhanced tissue regeneration in a full-thickness porcine skin defect model [10]. Age-related changes in large-vessel biomechanics were investigated in female C57BL/6NHsd mice with respect to neck vasculature and brain lipid biochemistry [11]. A tunable hyaluronan-elastin-like protein (HELP) hydrogel was engineered to independently control ligand presentation and matrix stress relaxation in glioblastoma cell studies [12].
