What is AMPK activation?

AMPK activation refers to the stimulation of AMP-activated protein kinase, a heterotrimeric serine/threonine kinase that acts as a cellular energy sensor. This page summarises research on pharmacological and physiologica

What is AMPK activation?

AMPK activation is the process by which AMP-activated protein kinase (AMPK), a heterotrimeric serine/threonine protein kinase composed of a catalytic α-subunit and regulatory β- and γ-subunits, is stimulated to phosphorylate downstream targets [8]. AMPK is often described as a cellular energy sensor because it responds to changes in adenine nucleotide levels and regulates metabolic pathways accordingly [4]. Activation can occur through allosteric binding of AMP or ADP to the γ-subunit, through phosphorylation of the α-subunit by upstream kinases such as LKB1 or CaMKKβ, or through pharmacological agents that directly or indirectly promote these events [8].

The research literature on AMPK activation centres on its role in the AMPK signalling pathway, which intersects with AKT signalling, GLUT4 translocation, NF-κB and JNK inflammatory cascades, and autophagy regulation [2][4].

Research Studies

Astaxanthin and granulosa-cell autophagy in PMOS-associated infertility

A triple-blind, placebo-controlled pilot randomised trial enrolled 50 women with polyendocrine metabolic ovarian syndrome (PMOS, formerly PCOS)-related anovulatory infertility between November 2023 and September 2024. Participants received astaxanthin (12 mg/day) or placebo. The study evaluated whether astaxanthin modulates autophagy-related signalling in granulosa cells and how these molecular effects relate to ovarian response [1]. The findings indicated that astaxanthin influenced autophagy activation in granulosa cells, though the pilot design limits generalisability [1].

Oxidative eustress and AMPK/AKT activation in skeletal muscle cells

In C2C12 myoblasts and myotubes, exposure to insulin and redox-modulating stimuli associated with oxidative eustress—including hydrogen peroxide, nitric oxide donors, and angiotensin II—was investigated for effects on AKT and AMPK phosphorylation, GLUT4 translocation, and functional glucose uptake. The study reported that moderate oxidation was associated with AMPK and AKT activation, GLUT4 translocation, and increased glucose uptake in skeletal muscle cells and fibres [2]. These findings are from in vitro and isolated fibre preparations, not from human trials.

Salicylate binding partners and AMPK dependency in renal epithelial cells

In the context of autosomal dominant polycystic kidney disease (ADPKD), salsalate has been shown to activate AMPK and ameliorate disease progression in vivo to a similar extent as tolvaptan. A targeted analysis of salicylate binding partners in renal epithelial cells revealed major AMPK dependency for these effects [3]. The work identified that salsalate affects multiple disease mechanisms, including metabolic reprogramming, cell proliferation, and inflammation, with AMPK playing a central role [3]. This research is at the cellular and animal model stage.

AMPK and macrophage crosstalk in diabetes

A review of AMPK and macrophage crosstalk in diabetes mellitus, especially type 2 diabetes (T2DM), examined how AMPK regulates macrophage polarization. In diabetes, adipose tissue macrophages adopt a pro-inflammatory M1 phenotype, secreting cytokines such as TNF-α, IL-6, and IL-1β that disrupt insulin signalling. AMPK suppresses M1 responses by inhibiting NF-κB and JNK signalling, activating CREB/SIRT1 pathways, and promoting oxidative metabolism [4]. This review synthesises preclinical and clinical observations but does not report new primary data.

Bitter melon compounds and AMPK activation in myotonic dystrophy type 1

Myotonic Dystrophy Type 1 (DM1) is characterised by repression of the AMPK pathway. A screen of 26 bitter melon-derived compounds (BMCs) in C2C12 myotubes identified BMC-25 as a potent AMPK activator. The study investigated the therapeutic potential of novel AMPK activators derived from Momordica charantia in a DM1 skeletal muscle phenotype, with effects observed in a sex-specific manner [5]. These findings are from cell-culture and animal models, not human trials.

Hepassocin and AMPK activation in liver senescence and regeneration

Hepassocin (HPS), a hepatokine with known hepatoprotective functions, was reported to prevent age-related liver senescence and facilitate liver regeneration by activating AMPK [7]. This research was conducted in animal models and identifies AMPK as a downstream mediator of HPS effects in the liver.

Pan-AMPK activator MK-8722 in human endothelial cells

A study comparing pharmacological AMPK activators in human endothelial cells reported that the pan-AMPK activator MK-8722 outperformed the AMPK activators SC4 and A-769662 [8]. AMPK regulates endothelial homeostasis and has emerged as a target for pharmacological activators in the treatment of endothelial dysfunction. This work is in vitro, using human endothelial cell cultures.

Palmitic acid and AMPK suppression in airway epithelial cells

In 16HBE airway epithelial cells, obesity-related palmitic acid was found to exacerbate IL-13-induced mucus phenotype via AMPK suppression [9]. The study examined how obesity-related factors interact with type 2 inflammatory signals to worsen airway epithelial mucus phenotype, a mechanism relevant to childhood asthma comorbidities. These findings are from cell-culture experiments.

Exercise preconditioning and AMPK-ACC signalling in myocardial ischemia/reperfusion

In C57BL/6J mice, six weeks of treadmill exercise preconditioning before myocardial ischemia/reperfusion injury was investigated for effects on ferroptosis. The study reported that exercise preconditioning activates AMPK-ACC signalling and attenuates ferroptosis [10]. This research is in an animal model and does not establish human outcomes.

Comparison of AMPK activators in human endothelial cells

CompoundRelative efficacy in human endothelial cells
MK-8722 (pan-AMPK activator)Outperformed SC4 and A-769662 [8]
SC4Less effective than MK-8722 [8]
A-769662Less effective than MK-8722 [8]
