Common misconceptions about AMPK activation
AMPK is a heterotrimeric serine/threonine protein kinase. This page addresses common misconceptions about its activation, drawing on laboratory and clinical evidence.
AMP-activated protein kinase (AMPK) is a heterotrimeric serine/threonine protein kinase consisting of a catalytic α-subunit and regulatory β- and γ-subunits [8]. It is also known as 5'-AMP-activated protein kinase. The evidence block does not provide sequence, formula, or molecular weight identifiers.
The work centres on the AMPK signalling pathway and its crosstalk with AKT, NF-κB, JNK, and CREB/SIRT1.
Research Studies
Astaxanthin and autophagy in granulosa cells from women with PMOS
A triple-blind, placebo-controlled pilot randomised trial enrolled 50 women with polyendocrine metabolic ovarian syndrome (PMOS)-related anovulatory infertility between November 2023 and September 2024. Participants received astaxanthin (12 mg/day) or placebo. The study evaluated whether astaxanthin modulates autophagy-related signalling in granulosa cells and how these molecular effects relate to ovarian response [1].
Oxidative eustress and AMPK/AKT activation in skeletal muscle cells
In C2C12 myoblasts and myotubes, AKT and AMPK phosphorylation were analysed after exposure to insulin and redox-modulating stimuli associated with oxidative eustress, including hydrogen peroxide, nitric oxide donors, and angiotensin II. GLUT4 translocation was assessed by quantitative immunocytochemistry. The study investigated the role of moderate oxidation in skeletal muscle glucose uptake, focusing on AKT/AMPK phosphorylation, GLUT4 translocation, and functional glucose uptake [2].
Salicylate binding partners and AMPK dependency in renal epithelial cells
In the context of autosomal dominant polycystic kidney disease (ADPKD), salsalate activation of AMPK ameliorates disease progression in vivo to a similar extent as tolvaptan. Salsalate affects multiple disease mechanisms, including metabolic reprogramming, cell proliferation, and inflammation. Targeted analysis of salicylate binding partners in renal epithelial cells revealed major AMPK dependency [3].
AMPK and macrophage crosstalk in diabetes
In diabetes mellitus, especially type 2 diabetes (T2DM), adipose tissue macrophages adopt a pro-inflammatory M1 phenotype, secreting cytokines including TNF-α, IL-6, and IL-1β that disrupt insulin signalling. AMPK suppresses M1 responses by inhibiting NF-κB and JNK signalling, activating CREB/SIRT1 pathways, and promoting oxidative metabolism. This review investigates the bidirectional interaction between AMPK signalling and macrophage function [4].
Bitter melon compound and AMPK activation in myotonic dystrophy type 1 skeletal muscle
Myotonic Dystrophy Type 1 (DM1) is characterised by repression of the AMPK pathway. A screen of 26 bitter melon-derived compounds (BMCs) in C2C12 myotubes identified BMC-25 as a potent AMPK activator. The study investigated the therapeutic potential of novel AMPK activators derived from Momordica charantia in a sex-specific manner [5].
TMZ plus 6-MP in recurrent glioblastoma
A clinical trial is evaluating the efficacy and safety of temozolomide (TMZ) plus 6-mercaptopurine (6-MP) in patients with recurrent glioblastoma. TMZ is an oral alkylating agent that traverses the blood-brain barrier. The trial description does not specify AMPK involvement [6].
Hepassocin and liver senescence
Hepassocin (HPS), a hepatokine, prevents age-related liver senescence and facilitates liver regeneration by activating AMPK [7].
Pan-AMPK activator MK-8722 in human endothelial cells
The pan-AMPK activator MK-8722 outperforms the AMPK activators SC4 and A-769662 in human endothelial cells. AMPK regulates endothelial homeostasis and has emerged as a target for pharmacological activators in the treatment of endothelial dysfunction [8].
Palmitic acid and AMPK suppression in airway epithelial cells
Obesity-related palmitic acid exacerbates IL-13-induced mucus phenotype in 16HBE airway epithelial cells via AMPK suppression. Mucus hypersecretion and obesity are prevalent comorbidities that independently worsen childhood asthma outcomes [9].
Exercise preconditioning and AMPK-ACC signalling in myocardial ischemia/reperfusion
In C57BL/6J mice, six weeks of treadmill exercise preconditioning activated AMPK-ACC signalling and attenuated myocardial ischemia/reperfusion-induced ferroptosis [10].
