What markers track AMPK activation?
A research-supply overview of the readouts used to track AMPK activation across cell, animal and human studies, including phosphorylation targets, downstream substrates and model-specific caveats.
What markers track AMPK activation?
AMP-activated protein kinase (AMPK) is a heterotrimeric serine/threonine protein kinase composed of a catalytic α-subunit and regulatory β- and γ-subunits [8]. In the research literature, AMPK activation is most commonly tracked by phosphorylation of the catalytic α-subunit at Thr172 (p-AMPKα Thr172), by phosphorylation of the downstream substrate acetyl-CoA carboxylase (p-ACC), and by functional readouts such as GLUT4 translocation and glucose uptake in muscle cells [2][10]. These markers are used across in vitro, animal and human study designs, but the evidence base for each marker differs by model and tissue.
The work centres on the AMPK signalling axis and its crosstalk with AKT, NF-κB, JNK and CREB/SIRT1 pathways in metabolic, inflammatory and endothelial contexts [2][4][8].
Research Studies
Oxidative eustress and AMPK/AKT phosphorylation in skeletal muscle cells
In C2C12 myoblasts and myotubes, moderate oxidation induced by hydrogen peroxide, nitric oxide donors and angiotensin II was associated with AKT and AMPK phosphorylation, GLUT4 translocation and functional glucose uptake [2]. The study analysed these readouts in cell culture and fibre preparations, positioning p-AMPK and p-AKT as markers of redox-sensitive glucose uptake signalling in skeletal muscle [2].
Salicylate binding partners and AMPK dependency in renal epithelial cells
Targeted analysis of salicylate binding partners in renal epithelial cells revealed major AMPK dependency [3]. In the context of autosomal dominant polycystic kidney disease (ADPKD), salsalate had previously been shown to activate AMPK and ameliorate disease progression in vivo to an extent similar to tolvaptan, affecting metabolic reprogramming, cell proliferation and inflammation [3]. The marker set in this work included AMPK activation status as a determinant of salicylate target engagement in renal epithelial cells [3].
AMPK and macrophage polarization in diabetes
A review of AMPK and macrophage crosstalk in diabetes reported that AMPK suppresses M1 responses by inhibiting NF-κB and JNK signalling, activating CREB/SIRT1 pathways and promoting oxidative metabolism [4]. In diabetes, adipose tissue macrophages adopt a pro-inflammatory M1 phenotype and secrete TNF-α, IL-6 and IL-1β, which disrupt insulin signalling [4]. Markers used to track AMPK activation in this context include p-AMPK, p-ACC and downstream inflammatory cytokine profiles in macrophage populations [4].
Bitter melon compounds and AMPK activation in DM1 skeletal muscle
A screen of 26 bitter melon-derived compounds (BMCs) in C2C12 myotubes identified BMC-25 as a potent AMPK activator [5]. In myotonic dystrophy type 1 (DM1), the AMPK pathway is repressed, and targeting AMPK has been proposed as a therapeutic avenue [5]. The study reported that BMC-25 ameliorated the DM1 skeletal muscle phenotype in a sex-specific manner in animal models [5]. Markers included AMPK activation status and skeletal muscle phenotype readouts [5].
Hepassocin and AMPK activation in liver senescence
Hepassocin (HPS), a hepatokine, was reported to prevent age-related liver senescence and facilitate liver regeneration by activating AMPK [7]. This reference-only evidence supports the use of p-AMPK and senescence markers in liver tissue, but the page does not build a section around it [7].
Pan-AMPK activator MK-8722 in human endothelial cells
The pan-AMPK activator MK-8722 outperformed the AMPK activators SC4 and A-769662 in human endothelial cells [8]. AMPK regulates endothelial homeostasis and has emerged as a target for pharmacological activators in endothelial dysfunction [8]. Markers in this work included AMPK activation status and endothelial functional readouts [8].
Palmitic acid and AMPK suppression in airway epithelial cells
Obesity-related palmitic acid exacerbated IL-13-induced mucus phenotype in 16HBE airway epithelial cells via AMPK suppression [9]. This reference-only evidence links AMPK suppression to mucus hypersecretion in the context of childhood asthma comorbidities [9]. Markers included AMPK activity status and mucus phenotype readouts [9].
Exercise preconditioning and AMPK-ACC signalling in myocardial ischemia/reperfusion
In C57BL/6J mice, six weeks of treadmill exercise preconditioning activated AMPK-ACC signalling and attenuated myocardial ischemia/reperfusion-induced ferroptosis [10]. This reference-only evidence supports p-AMPK and p-ACC as markers of exercise-induced cardioprotection in animal models [10].
Astaxanthin and autophagy signalling in granulosa cells
A triple-blind, placebo-controlled pilot randomised trial in women with PMOS-related anovulatory infertility evaluated whether astaxanthin (12 mg/day) modulates autophagy-related signalling in granulosa cells [1]. The study enrolled 50 women between November 2023 and September 2024 [1]. Markers included autophagy-related signalling readouts in granulosa cells and ovarian response measures [1].
